Danneel Ackles is set to guest star in “Supernatural,” Variety has learned exclusively.

Ackles is set to recur as “Sister Jo” in the later part of the 13th season of the CW’s hit demon hunting drama. Sister Jo is described as “a well-known faith healer whose good works attract the attention of someone very, very bad: Lucifer (Mark Pellegrino).” The question she will have to face is whether Lucifer will kill her — or if he has bigger plans for her.

Her first appearance will be in episode 13, “Devil’s Bargain.”

Ackles joins a list of guest stars for this record season that include Christian Keyes, Jeffrey Vincent Parise, Keith Szarabajka, Yadira Guevara-Prip and Clark Backo, as well as the returns of David Haydn-Jones, Osric Chau, Lorette Devine, Briana Buckmaster, Kathryn Newton, Katherine Ramdeen and Kim Rhodes.

Ackles, who is married to “Supernatural” star Jensen Ackles is best known for starring in the cult classic “Harold & Kumar” film franchise, in addition to her role as Rachel Gatina on the long-running WB series “One Tree Hill.”

She previously appeared with her husband in the 2007 independent film “Ten Inch Hero,” and in 2014 she worked with “Supernatural” star Misha Collins in his digital short “TSA America: Suspicious Bulges.”

Another Ackles is joining the Supernatural fun.

EW has confirmed that Danneel Ackles will recur in the back-half of Supernatural‘s 13th season. Ackles, who’s married to series star Jensen Ackles, will play a character named Sister Jo.

According to Variety, who first reported the casting, Sister Jo is a faith healer who catches the eye of none other than Lucifer. And as fans know, once you’ve piqued Lucifer’s interest, well, your life becomes a whole lot more complicated.

So far in season 13, Lucifer’s main mission has been to find his son, though at this point, you can add “stop Michael” to his to-do list as well. There’s no word on whether Sister Jo will tie into either of those schemes (or whether Ackles will get to share any screen time with her husband).

Ackles, best known for her work on One Tree Hill, will first appear in episode 13, titled “Devil’s Bargain.”

Supernatural airs Thursdays at 8 p.m. ET on The CW.

When you think about it, it's a little nuts it took this long for Supernatural to cast Jensen Ackles' wife, Danneel Ackles, in a guest role, but it's finally happening!

According to Variety, Danneel Ackles has been given a guest role in Season 13 as a new character, Sister Jo. Sister Jo is described as "a well-known faith healer whose good works attract the attention of someone very, very bad: Lucifer (Mark Pellegrino)."

A faith healer and the devil? Throw the phrase "walk into a bar" in there, and there's endless amounts of jokes for Sam (Jared Padalecki) and Dean to make.

Danneel Ackles is no stranger to The CW, having spent four years playing Rachel Gatina on One Tree Hill. She's appeared on CSI, NCIS and Charmed, as well as in several films. This actually won't be the first time she's acted opposite Jensen Ackles, seeing as they filmed Ten Inch Hero together back in 2007.

The couple married in May of 2010 and have three children together — twins, Zeppelin and Arrow and their oldest child, Justice.

What's most interesting is that Variety reports she'll make her first appearance in Episode 13, "Devil's Bargain." First appearance implies that she'll be making more than one, turning this little guest role into a potential guest arc.

We can be the only ones dying to see Jensen and Danneel act opposite each other for multiple episodes this season, right?

Between the fog and the film crews, Cloverdale is feeling pretty Supernatural today.

The cast and crew of the hit American fantasy-horror TV series Supernatural is filming scenes for Season 13’s episode “Devil’s Bargain” on Monday and Tuesday along 176 Street in the downtown core.

Crews were on site last night to take down Christmas lights and wreaths that would have been in the shot. They hung flower baskets instead.

Got in to work today to find that film crews had replaced all the Christmas decorations with flower baskets. It is very summery (minus the temperature, fog, and everyone wearing winter clothing).

The popular TV series follows the Winchester brothers as they hunt the things that goes bump in the night. The two brothers, played by actors Jensen Ackles and Jared Padalecki, face off against supernatural creatures, from gods to monsters and everything in between.

On Monday morning, an actress could be seen rollerblading down the sidewalk past Elena’s Country Kitchen, narrowly missing pedestrians until she came to a stop by crashing into an actor holding a cake.

The series, which is filmed in Vancouver and the Lower Mainland, will be filming on site today in front of Elena’s Country Kitchen until 11:30 a.m., and will then shoot from noon to 3:30 p.m. in the alleyway behind Nana’s Antiques.

The crews will be back on Tuesday to film additional scenes along 176 Street.

The crew constructed a faux burger shop and a gazebo in Hawthorne Square at 5755 176 Street and on Tuesday morning, Mark Pellegrino, who plays Supernatural’s Lucifer, was seen filming close shots.

Later in the afternoon, Jensen Ackles (Dean Winchester), Jared Padalecki (Sam Winchester) and Misha Collins (Castiel) were seen on set rehearsing takes.

A small group of fans gathered on the street opposite from filming to view the action.

The filming is expected to move to 5780 176A Street to film exterior shots in the afternoon before wrapping at 4:30 p.m. Crew will return on Wednesday to restore the Christmas decorations on 176 Street, which they took down to film this week.

Supernatural is filming the Devil’s Bargain, the thirteenth episode of its thirteenth season. And it’s a big deal. Danneel Ackles (One Tree Hill) guest-stars as faith healer Sister Jo, who comes to the attention of Supernatural’s Big Bad Lucifer (Mark Pellegrino). Her character will recur in the back half of the season.

ENTERTAINMENT WEEKLY: After 13 years, what made you finally want to join in on the fun?
DANNEEL ACKLES: Mostly the character. I haven’t acted in a couple years and Eugenie [Ross-Leming], who is Bob Singer’s wife and one of the executive producers and writers on the show, wrote this character with her partner, Brad [Buckner], and she called me and she pitched it. [Sister Jo] is such a badass. I was like, “Yeah, this is what I want to do.” I didn’t want to go on Supernatural and be a love interest of the week or have the guys kill me in some horrific way. But this character, she’s just a badass. And where else can you go and bring your kids to work and hand them off to A camera while you’re shooting? So it was a no-brainer for me.

Did they write the character with you in mind?
That’s what I’ve been told. They were like, “She’s basically you so have fun with it.”

All we know about Jo so far is that she’s a faith healer. How would you describe her more generally as a person?
She is a businesswoman. She is a legitimate faith healer and a very powerful faith healer, and she loves doing it, but she is a businesswoman first. So she is always looking for: “How much money can I charge this person in this town?” It’s all about business for her.

We know she finds herself in a deal with Lucifer. Is this something she enters into willingly?
She’s attracted to power, and she, herself, is a very powerful, big personality, and so she initially is very attracted to him because of that. She’s like, “Okay, I’m a powerful person, you’re a powerful person, let’s see if we can work something out that benefits us.” So it’s not really like she’s a good girl or a bad girl; she’s about what’s going to be best for Sister Jo.

What are her thoughts on the Winchesters?
It goes back to her not really being good or bad. She’s just going to do whatever works for her. She’s approached by Lucifer and then the Winchesters are attracted to Lucifer — obviously they’re looking for him — so they come across Sister Jo, and she is the type of person that could have a lot of deals going at once. They’re looking for Lucifer and she’s like, “Well, I can help with that.” So she’s not really loyal to anyone in particular but herself. She is numero uno.

This is obviously a set and crew you’ve been around for years, but this is your first time acting on this set. Did anything surprise you about the experience?
My first scene that I did — we shoot out of order — the first scene was with Jared [Padalecki] and Jensen [Ackles] and Misha [Collins]. I hadn’t worked in like four years and the first thing I had to do was walk into a scene with those goofballs who were looking at this like it was the most surreal thing. The four of us are standing there, all looking at each other like, “This is a weird dream.” There was a lot of laughing, which was great. I was nervous that that was the first scene, but looking back on it now, it was just a great way to break the ice and get in. And 40 takes later, we were actually over the fact that it was the four of us and we got on with it. [Laughs] But it was a lot of fun.

So fans can look forward to the gag reel.
Oh, there’s a great moment there — a really good one, courtesy of me, take three. There might’ve been some falling. Something to look out for. [Laughs]

“She’s not really, ‘I’m an evil person’ or ‘I’m a good person,'” the actress tells TVLine. Instead, Sister Jo’s line of thinking is more along the lines of, “‘I’m just going to do whatever gets me a little bit higher up the ladder.’ So if she can make a deal with Lucifer that day, that’s great for her, [and] she’s going to do that. If she needs to help the Winchesters another day, then that works.”

Below, Danneel Ackles reveals how the “badass” role came to be hers and previews Sister Jo’s agenda.

TVLINE | Has this been something that you’ve wanted to do for a while, to be on the show?
Not really. Back when the show first started two million years ago, I was out on One Tree Hill, and [Jensen] was up in Canada. So we were both very busy. Then when we started having kids and things slowed down, Supernatural was already on its way, and I thought, “That’s his thing.” But now, Eugenie [Ross-Leming] and Brad [Buckner], who are executive producers of the show, came up with this character and pitched it to me, and I thought, “Yeah! This is it. This is the reason to do it.” She’s a badass female character who just knows what she wants, and they kind of wrote it with me in mind, they say. So I was just really attracted to it, and wanted to give it a shot.

TVLINE | Besides being a badass, what else defines her?
They kind of pitched her as a business woman. She’s in it for herself — not in a negative way. Not in a “I’ll step on whoever I have to make things great for me” [way]. More in the way of, “You know what? That sounds good. I like that today. And that sounds great. I like that. That works for me, and that’ll help.” So she’s a business woman and a very powerful faith healer.

TVLINE | So the faith healing is for real? It’s not part of some scheme?
No. It’s the real deal. She is an extremely powerful faith healer… so powerful that she attracts Lucifer’s attention. That’s what initially starts all this. And wherever Lucifer goes, the Winchesters go. That’s how we all end up in this little pocket together.

TVLINE | How aware is she of who Lucifer is and the supernatural world?
I don’t know at first if she is. She’s just very aware of how powerful he is. And like I said earlier, she is really drawn to powerful people, and people that can help her move forward in her life. So that attracts her to him. She’s like, “He can help me.” And he’s very charming.

TVLINE | The Winchesters have a very long and complicated history with faith, dating back to Season 1. What do they make of Sister Jo and her abilities?
At first, I think they’re a little skeptical. But they’ve seen a lot in their day. They don’t really come after her, because they’re like, “Well, maybe she can help us.” They kind of leave her alone because she has some qualities that they need to use. Everybody’s using everybody!

TVLINE | And what are her first impressions of Dean and Sam?
I don’t think she really cares too much about them. They ask for assistance, she works a deal with them because she’s this deal-making business woman, and that’s that.

TVLINE | Jared [Padalecki] and Jensen have a reputation for hazing newcomers. Did they try to pull any pranks on you?
You know, I think they would have, except I did such a good job of doing it to myself that, no, they didn’t really have to. They let me concentrate. I only had a couple scenes with them. So I think had there been more opportunity… I lucked out. One of our scenes was first in the day, and the other one was really quick, and it was right before they had a ton of work to do. And that’s the key with those two: If you get them when they’re a little slaphappy at the end of the night, that’s when you’re asking for trouble.

TVLINE | You obviously have a unique insider/outsider perspective on the show and are very familiar with it. When you got to set to shoot, did anything surprise you about the experience?
I was surprised how short Jensen was and [how] tall Jared was. I’m just kidding. [Laughs] No, it was pretty professional. The first two or three scenes we did, we all kept having these surreal moments. I believe there was one time I called Jared “Jared” in the take. No, we all buckled down, and it was pretty good.

What was it about this time in the show’s history and your own life that made it the right moment to come on “Supernatural”?

The show has a huge fan based and they’re all on social media, and somebody pointed out to me that in 2006, on Twitter, I said that if “Supernatural” goes to Season 13, that’s when I’m going to guest star. And they screenshotted it and sent it back to me, so I guess the universe was like, “Yep, it’s time.” But I think it was just more the offer because before there had been some talk of going on and doing something like a demon or a sexy-something, and this particular role really interested me, and I thought it was good, and I don’t have to play a love interest of Jensen’s, so it works.

Is this marking a turning point for you, where you want to come back to acting?

You know, I would love to, but we have twin one-year-olds and a four-year-old and we just had another baby, which is our brewery we just opened about 30 days ago. So I would love to [and] if the opportunity came up, and it worked like this, great. This is fun, and literally we had all our kids in the trailer, and I was just running back and forth, and [Jensen] was the babysitter. It just worked.

How different is Sister Jo from previous faith healers the show has introduced?

She’s an extremely powerful faith healer — powerful enough that Lucifer finds out about her and is intrigued. He wants to elicit her help. She’s powerful enough that she’s able to negotiate with him, and that makes her different.

Where does Sister Jo fit in when it comes to the female characters on the show? Is there anyone you would compare her to when it comes to the type of strength or abilities she exhibits?

She’s definitely in line with all of the [women] in the “Wayward Sisters” spinoff. All of those female characters are so strong, and “Supernatural” is great about writing strong, female characters, and I feel like the majority of them hold their own. So I feel like in that she fits in with how “Supernatural” writes women.

What is it about her meeting with Lucifer that either inspires her to want to make a deal or forces her to have to make a deal?

Lucifer in “Supernatural” is very charming and kind of sexy in an offbeat way. She’s a very strong woman, and I think that he intrigues her. She’s attracted to him and his power. I think they kind of like that about each other.

How quickly do things go south?

I don’t know that yet. We have talked about doing another episode sometime before the end of the year, so we might find out then how it all ends up, but as of right now, I don’t know.

Obviously she encounters the Winchesters pretty quickly — what does the dynamic look like when she does?

It’s good. She ends up helping them in a way. Because Lucifer has found her, that then attracts the Winchesters. That’s how they end up meeting, and she ends up helping them track Lucifer.

Is she genuinely helping them find him, or is she more aligned with Lucifer?

I think we can think about her as more aligned with herself. She’s a faith healer, but also a businesswoman. She thinks about herself first, and if she can help out somebody along the way, great, but she is No.1.

In the past, the show has gotten very meta, winking at actors’ past works or relationships. Does that happen with you — little nods to either your marriage to Jensen or previous collaboration in “Ten Inch Hero”?

Good God, no! At least not “Ten Inch Hero.” But no, I think they kept it pretty clean.

Do you feel like that’s better for the character because it just lets her live in the world of the show on her own?

Absolutely. I think it worked really well with Jared [Padalecki’s] wife Gen — I think it was great when they did “The French Mistake;” it played perfectly because she was Ruby before she was his wife. And I think that’s kind of been done, so they wanted this to be cleaner and she can be a standalone character.

How was it stepping onto the set to work with all of these people — cast and crew — that you’ve developed such close relationships with through Jensen through the years?

Very surreal. It was great. Most of my scenes were with Mark Pellegrino, not really the guys too much, though we had a few things together. Jensen’s so supportive and he’s so sweet, and then we get to set and he’s like the big prankster — laughing, making faces at me behind the camera. And I’m like, “Please take it easy on me. I haven’t acted in, like, six years. I need professionalism.” So we had, I think, just the right amount of interaction, but it was fun — it was really fun.

The pranks come up all the time, but were you off-limits for any kind of hazing?

I think so. Nobody really pranked me except just acted really goofy during all of our scenes that were supposed to be really serious.

Aside from the personal relationship, you also worked with Jensen years ago. How did it feel to work with him again now?

Not that call sheet numbers matter, but when we did “Ten Inch Hero,” I was like No. 4 and he was like No. 27, and I think that dynamic has totally changed. Now he’s directed and Jensen knows “Supernatural” from behind the camera and in front of the camera, and all of the little nuances that make the show so great, so I just really leaned on him more than ever. “What do I do here? How is my pacing?” He is the show, and he knows that, so I just really leaned on him for direction, so if everyone hates it, it’s all his fault.

ET: How much of a discussion was it on your end in regards to the perfect time for you to appear on Supernatural?

Danneel Ackles: I was reminded of this in the Twitter-verse. Fans were always like, "Are you going to be on Supernatural? When are you going to be on Supernatural?" and in 2006, I was like, "Well, in season 13, I'm going to get there on Supernatural." A fan found that and has now retweeted it and reposted it and I was like, "Are you kidding me? I said that?" I guess it was the universe's way of making it come to fruition.

It has been a significant period of time, over a decade, since the show has been on. Was it a conscious decision on your part to keep your distance from it?

In the beginning, I was working on different shows throughout and [Jensen] just kept staying on the same one. (Laughs.) We were working so much on different things that it really didn't come up. And this particular character, [Sister Jo], was written by Eugenie Ross-Leming and Brad Buckner, who are executive producers on the show, and Eugenie's husband is Bob Singer, who is also very prominent on the show. Eugenie and Brad pitched the character to me and she's just a really strong, female badass and I was like, yes, I want to play that. It wasn't playing me, which, I think worked great in [the 2011 episode] "The French Mistake" for Gen[evieve Padalecki, who played herself]; she was Ruby before she was Genevieve Padalecki. I think that worked really well and fans absolutely loved the episode. For Jensen and I, this felt better. He read it, I read it, this is an awesome character and I have to do it.

Let's talk a little bit about your character, Sister Jo, who is a faith healer and attracts the attention of Lucifer, which is usually a bad thing but you never know. What drew you into Sister Jo?

She does come into contact with Lucifer. They're both very drawn to each other. She's a faith healer. She's extremely powerful, which is something we know he is very attracted to. The one thing about Sister Jo is that she is a very smart businesswoman. She's not a bad girl or a good girl, she's one of these types who is driven and "OK, Lucifer wants to make a deal with me. This works for me. I'm going to do that." Very matter of fact. Very in it to see who can help her climb the ladder of life quicker. In this episode, she does deals with Lucifer, she does deals with the Winchesters. As long as you're helping No. 1, Sister Jo, you're good in her book.

So she's someone who operates more in the gray area.

Exactly.

You mentioned sharing scenes with the Winchester boys. You worked with Jensen more than 10 years ago on the 2007 film, Ten-Inch Hero. What was this experience like working together?

Over a decade ago! That was a very different experience. Aside from both of us having very different bodies, our bodies have [changed since] -- I would love to go back to that. (Laughs.) But this time, I'm handing babies off. I was like, "Here's this baby!," because we have 1-year-old twins and a 4-year-old. We're running back and forth from trailers. It was crazy just on that aspect alone. I do know how lucky I am because there's no other show where I can do a scene and hand off a baby to the props girl, Robin, who we love. It was really fun. Good times.

What can you tease about Sister Jo's dynamic with the brothers?

It's the same dynamic as when she first meets Lucifer. It's the same kind of situation for her where they ask her for help and she's like, "OK, what can you do for me?" That's her dynamic. She uses what she has at the moment to get what she needs.

You played a memorable bad girl, Rachel, on One Tree Hill. Do you think Sister Jo has traces of Rachel in her DNA?

I think there's a little Rachel in Sister Jo, definitely the type that is driven because Rachel was very driven and very much in it to win it. If you could help her, then you're a friend. Sister Jo is not very judgmental. She's willing to overlook mistakes; if she thinks you can help her in this moment... Ehh, so you're Lucifer? Big deal.

Is there a trait of Jo's you wish you had in real life?

I wish I was as fearless as she was. If I walked into a room and the devil was there, I would be out quicker than... (Laughs.) I don't even go to haunted houses! It would probably be her fearlessness. Or, maybe I should be a faith healer. I'd actually really want to be that too. That would be really cool. The director of this episode was Eduardo Sanchez, who did The Blair Witch Project. I couldn't even ask him about it. I didn't want to talk about that movie with him because that movie scarred me when I was younger.

What do you want fans to know about your arc now that you're finally in the Supernatural universe?

I'm going to be doing another episode before the show ends this season and I'm super excited about it. I really love the character and I hope that she can continue on. If they do decide to kill her, I hope it's not some really horrible death. (Laughs.) Or hopefully it's not at the hands of Dean Winchester.

Fans would riot.

Well, you remember what they did to Jared [Padalecki]'s wife. Wasn't it that Jared held her while Jensen stabbed her? [In the season six finale, Ruby was fatally stabbed by her own knife by Dean while Sam held her in his arms.] Please don't let that happen to me! Jared's like, that's what he's rooting for.

So excited for Danneel to be on Supernatural! What do you know about her first episode? — Crystal
The episode was directed by Eduardo Sanchez, who helmed The Blair Witch Project, so you wanna take a guess at what that means? Actually, it might not be quite what you’re imagining. According to Danneel Ackles, the episode’s a bit “scarier” than usual, but nothing out of the ordinary. “It was pretty straightforward,” she says “There’s some stuff coming down the line, like the Scooby-Doo episode, so this one pretty much brings us back to being a very straightforward Supernatural episode.” But there is one question Danneel wants to clear up: “There were no references to the fact that Jensen and I know each other. Some people were asking if we were going to do something like that.”
